Effect of Plant Hormones on Tobacco Mosaic Virus Concentration in Tobacco Tissue Culture

چکیده
Tobacco mosaic virus (TMV) concentrations in tobacco callus tissues grown on the media containing cu-naphthaleneacetic acid (NAA) and kinetin at different concentrations were assayed periodically during successive cultures. Tobacco callus originally raised from the TMV-infected tobacco stem became green and compact when serially cultured on the medium containing lower concentration (0.01-o. 1 mg/l) of NAA or higher concentration (2mg/l) of kinetin. Translucent and soft callus was obtained at higher concentration (l-40 mg/l) of NAA or lower concentration (O-O. 2 “g/l) of kinetin. Microscopic observations of the thin sectioned callus tissues revieled that the green colored compact tissue was composed of the closely associated small cells and many tracheid-like structures, while in the translucent and soft tissue, large cells were roughly arranged showing less association and no tracheidlike structure was found. TMV was maintained at high concentration in the compact callus during successive cultures, however, it was no longer detected beyond the third generation (90 days) in the translucent soft callus grown under the optimum hormone balances. Since both hormones do not have direct inactivation effect on TMV, the results suggest that TMV multiplication in the callus tissue closely corresponds with the cell arrangement of the tissue, which is considerably affected by hormone balances in the media.

Tomato and Tobacco Phytoene Desaturase Gene Silencing by Virus-Induced Gene Silencing (VIGS) Technique
Background and Aims: Virus-Induced Gene Silencing (VIGS) is a virus vector technology that exploits antiviral defense mechanism. By infecting plants with recombinant viruses containing host genes inserted in the viral genome, VIGS achieves the RNA silencing process. The purpose of this study was to investigate the possibility of tomato (Lycopersicon esculentum Mill.) and tobacco (Nicotiana be...
متن کاملStudy on Genetic Diversity of Terminal Fragment Sequence of Isolated Persian Tobacco Mosaic Virus
Tobacco mosaic virus (TMV) is one of the devastating plant viruses in the world that infects more than 200 plant species. Movement protein plays a supportive role in the movement of other plant viruses, and viral coat protein is highly expressed in infected plants and affects replication and movements of TMV. In order to investigate genetic variation in the terminal fragment sequence in Iranian...
متن کاملA systemic increase in the recombination frequency upon local infection of Arabidopsis thaliana plants with oilseed rape mosaic virus depends on plant age, the initial inoculum concentration and the time for virus replication
In the past, we showed that local infection of tobacco leaves with either tobacco mosaic virus or oilseed rape mosaic virus (ORMV) resulted in a systemic increase in the homologous recombination frequency (HRF). Later on, we showed that a similar phenomenon occurs in Arabidopsis thaliana plants infected with ORMV. Here, we tested whether the time of removing the infected leaves as well as viral...
متن کاملLocal and systemic spread of tobacco mosaic virus in transgenic tobacco.
Expression of a chimeric gene encoding the coat protein (CP) of tobacco mosaic virus (TMV) in transgenic tobacco plants confers resistance to infection by TMV. We investigated the spread of TMV within the inoculated leaf and throughout the plant following inoculation. Plants that expressed the CP gene [CP(+)] and those that did not [CP(-)] accumulated equivalent amounts of virus in the inoculat...
